摘要: A co-expressing system of DsbA-DsbA was suggested for the first time to enhance the soluble expression of human trypsin-1. As a control, leaderless DsbA chaperone was also co-expressed with human trypsin-1. Vectors pET39b-trypsin and pET28a-DsbA-DsbA -trypsin with the above two DsbA fusion tag were constructed. The strain with vector pET39b-trypsin expressed fusion protein DsbA-trypsin in form of inclusion bodies. While in BL21 (DE3) strain with vector pET28a-DsbA-DsbA -trypsin, the soluble expression of trypsin fusion protein was achieved. Under the optimized expression conditions, the soluble fraction accounted for about 49.43% of total DsbA-DsbA -trypsin proteins in crude supernatant. The purification yield was 4.15% by nickel chelating chromatography and 3.3 mg activated trypsin with a purity of 88.68% was obtained from 1 L LB broth. To detect the possible functions of DsbA series chaperons in trypsin fusion protein, we analyzed the primary three-dimensional structure of fusion proteins, mainly focusing on the compatibleness between trypsin and fusion chaperons. The results suggested that (1) besides the primary function in periplasm, leaderless DsbA or DsbA may also act as a signal sequences-like leader targeted to periplasm that partly relieved the pressure from fusion protein overexpression and inclusion body formation, and (2) as there was significant soluble expression of DsbA-DsbA -trypsin compared with DsbA-trypsin, DsbA may function as charge or hydrophobic balance in recombinant protein DsbA-DsbA -trypsin.

In this study, the distributions of the absolute molecular weight and intrinsic viscosity of SMP polysaccharides from an AnMBR were effectively determined by a high performance size exclusion chromatography (HPSEC) that was coupled with the refractive index (RI), diode array UV (DAUV), right and low angle light scattering (LS), and viscometer (Vis) detectors. Based on the tetra-detector HPSEC determined absolute molecular weights and intrinsic viscosity, a universal calibration relationship for the SMP polysaccharides was developed and the molecular conformations, average molecular weights, and hydrodynamic sizes of the SMP polysaccharides were also explored. Two factors which can be derived from the tetra-detector HPSEC analysis were proposed for the characterization of the viscous and osmotic pressure properties of the SMP polysaccharides. In addition, it was also extrapolated how to analyze the resistance characteristics of the concentration polarization layers formed in membrane filtration based on the molecular properties determined by the tetra-detector HPSEC analysis.

Characteristics of mass distributions of aerosol particle and its inorganic water-soluble ions in summer

摘要: Agricultural activity is one of the most important sources of aerosol particles. To understand the mass distribution and sources of aerosol particles and their inorganic water-soluble ions in a suburb farmland of Beijing, particle samples were collected using a microorifice uniform deposit impactor (MOUDI) in the summer of 2004 in a suburb vegetable field. The distribution of the particles and their inorganic water-soluble ions in the diameter range of 0.18 18 mm were measured. The dominant fine particle ions were SO, NO, and NH. The association of day-to-day variation of the concentration of these ions with temperature, humidity, and solar radiation suggested that they are formed by the reaction of NH3 released from the vegetable field with the acid species produced from photochemical reactions. Fine particle K is likely from vegetation emission and biomass burning. Coarse particles like Ca, Mg, NO, and SO are suggested to come from the mechanical process by which the soil particle entered the atmosphere, and from the reaction of the acid species at the surface of the soil particle. The results show that fertilizer and soil are important factors determining the aerosol particle over agricultural fields, and vegetable fields in suburban Beijing contribute significantly to the aerosol particle.

Synchronous observation of rising soluble bubble through quiescent solution

摘要: An experimental method using computer image processing technology (CIPT) was proposed to observe and investigate the velocity, deformation, heat and mass transfer, etc. of a rising soluble gas (CO) bubble through a quiescent hot water. A model was set up to describe the behavior of the bubble in a visual experimental system in which a high-speed camera rose instantaneously with the movement of the bubble. A series of trajectory videos about the bubble were recorded by a computer linked to the camera. The trajectory, volume changes and rate of mass transfer of the bubble were obtained by the CIPT. It is found that the single bubble follows a rolling trajectory at the initial stage when there is mass transfer. With the volume decreasing, the disturbed behavior of the bubble becomes tempered. When the rising velocity of the bubble reaches the maximum, the velocity is nearly at a constant. The experimental and analysis results show that this method is useful for the research on the mass transfer and the movement of rising bubbles in liquid.

Formation of disinfection byproducts from accumulated soluble products of oleaginous microalga after

摘要: When microalgae are simultaneously applied for wastewater treatment and lipid production, soluble algal products (SAP) should be paid much attention, as they are important precursors for formation of disinfection byproducts (DBPs), which have potential risks for human health. sp. HQ is an oleaginous microalga that can generate SAP during growth, especially in the exponential phase. This study investigated the contribution of SAP from sp. HQ to DBP formation after chlorination. The predominant DBP precursors from SAP were identified with the 3D excitation-emission matrix fluorescence. After chlorination, a significant reduction was observed in the fluorescence intensity of five specific fluorescence regions, particularly aromatic proteins and soluble microbial by-product-like regions, accompanied with slight shifting of the peak. The produced DBPs were demonstrated to include trihalomethanes and haloacetic acids. As the algal cultivation time was extended in wastewater, the accumulated SAP strengthened the formation of DBPs. The trend for DBP formation was as follows: chloroform>dichloroacetic acid>trichloroacetic acid.

Evaluation of factors influencing soluble microbial product in submerged MBR through hybrid ASM model

摘要: In this study, a mathematical model was established to predict the formation of the soluble microbial product (SMP) in a submerged membrane bioreactor. The developed model was calibrated under the reference condition. Simulation results were in good agreement with the measured results under the reference condition. The calibrated model was then used in the scenario studies to evaluate the effect of three chosen operating parameters: hydraulic retention time (HRT), dissolved oxygen concentration, and sludge retention time (SRT). Simulation results revealed that the SMP dominated the soluble organic substances in the supernatant. The scenario studies also revealed that the HRT can be decreased to 1 h without deteriorating the effluent quality; dissolved oxygen concentration in the reactor can be kept at 2-3 mg/L to maintain the effluent quality, reduce the content of SMP, and minimize operating costs; the optimal SRT can be controlled to 10-15 d to achieve complete nitrification process, less membrane fouling potential, and acceptable organic removal efficiency.

Lithium-ion modified cellulose as a water-soluble binder for Li-O battery

摘要: An environment-friendly, water-soluble, and cellulose based binder (lithium carboxymethyl cellulose, CMC-Li) was successfully synthesized by using Li+ to replace Na+ in the commercial sodium carboxymethyl cellulose (CMC-Na). Li-O2 batteries based on the CMC-Li binder present enhanced discharge specific capacities (11151 mA·h/g at 100 mA/g) and a superior cycling stability (100 cycles at 200 mA/g) compared with those based on the CMC-Na binder. The enhanced performance may originate from the electrochemical stability of the CMC-Li binder and the ion-conductive nature of CMC-Li, which promotes the diffusion of Li+ in the cathode and consequently retards the increase of charge transfer resistance of the cathode during cycling. The results show that the water-soluble CMC-Li binder can be a green substitute for poly(vinylidene fluoride) (PVDF) binder based on organic solvent in the lithium oxygen batteries (LOBs).

Buxu Tongyu Granule Alleviates Myocardial Ischemia by Activating Vascular Smooth Muscle Cell Soluble

摘要: Myocardial ischemia is a serious threat to human health, and vascular dysfunction is its main cause. Buxu Tongyu Granule (BXTY) is an effective traditional Chinese medicine (TCM) for treating myocardial ischemia. However, the underlying mechanism of BXTY is still unclear. In this study, we demonstrate that BXTY ameliorates myocardial ischemia by activating the soluble guanylate cyclase (sGC)–3′,5′-cyclic guanosine monophosphate (cGMP)–protein kinase G (PKG) signaling pathway in vascular smooth muscle cells (VSMCs) to dilate the arteries. BXTY was given by gavage for ten consecutive days before establishing an animal model of acute myocardial ischemia in mice via the intraperitoneal injection of pituitrin. The results showed that BXTY alleviated the symptoms of myocardial ischemia induced by pituitrin in mice, including electrocardiogram abnormalities and changes in plasma enzymes. In addition, BXTY dilated pre-constricted blood vessels and inhibited the vasoconstriction of the superior mesenteric artery in a dose-dependent but endothelial-independent manner. These effects were eliminated by pre-incubating vascular rings with the sGC inhibitors NS 2028 or ODQ, or with the PKG inhibitor KT 5823. Moreover, BXTY increased the protein expression of sGC-β1 and the intracellular second messenger cGMP level in mouse aortic vascular smooth muscle cells (MOVAs). NS 2028 or ODQ reversed these effects of BXTY. The expression level of the cGMP downstream effector protein PKG-1 increased after treating MOVAs with BXTY. NS 2028, ODQ, or KT 5823 also reversed this effect of BXTY. In conclusion, BXTY can improve the symptoms of acute myocardial ischemia in mice, and activating the sGC–cGMP–PKG pathway in VSMCs to induce vasodilation is its key pharmacodynamic mechanism.

Use of gamma-irradiation pretreatment for enhancement of anaerobic digestibility of sewage sludge

摘要: The effects of ?-irradiation pretreatment on anaerobic digestibility of sewage sludge was investigated in this paper. Parameters like solid components, soluble components, and biogas production of anaerobic digestion experiment for sewage sludge were measured. The values of these parameters were compared before and after ?-irradiation pretreatment. Total solid (TS), volatile solid (VS), suspended solid (SS), volatile suspended solid (VSS), and average floc size of samples decreased after ?-irradiation treatment. Besides, floc size distribution of sewage sludge shifted from 80–100 ?m to 0–40 ?m after ?-irradiation treatment at the doses from 0 to 30 kGy, which indicated the disintegration of sewage sludge. Moreover, microbe cells of sewage sludge were ruptured by ?-irradiation treatment, which resulted in the release of cytoplasm and increase of soluble chemical oxygen demand (SCOD). Both sludge disintegration and microbe cells rupture enhanced the subsequent anaerobic digestion process, which was demonstrated by the increase of accumulated biogas production. Compared with digesters fed with none irradiated sludge, the accumulated biogas production increased 44, 98, and 178 mL for digesters fed sludge irradiated at 2.48, 6.51, and 11.24 kGy, respectively. The results indicated that ?-irradiation pretreatment could effectively enhance anaerobic digestibility of sewage sludge, and correspondingly, could accelerate hydrolysis process, shorten sludge retention time of sludge anaerobic digestion process.

Plasma soluble C-type lectin-like receptor-2 is associated with the risk of coronary artery disease

摘要: Accumulating evidence suggests that C-type lectin-like receptor-2 (CLEC-2) plays an important role in atherothrombosis. In this case-control study, we investigated the association between CLEC-2 and incidence of coronary artery disease (CAD). A total of 216 patients, including 14 cases of stable angina pectoris (SAP, non-ACS) and 202 cases of acute coronary syndrome (ACS), and 89 non-CAD control subjects were enrolled. Plasma levels of soluble CLEC-2 (sCLEC-2) were measured using the enzyme-linked immunosorbent assay (ELISA). Compared with the control group (65.69 (55.36–143.22) pg/mL), the plasma levels of sCLEC-2 were significantly increased in patients with CAD (133.67 (88.76–220.09) pg/mL) and ACS (134.16 (88.88–225.81) pg/mL). The multivariate adjusted odds ratios (95% confidence interval) of CAD reached 2.01 (1.52–2.66) ( <0.001) for each 1-quartile increase in sCLEC-2. Restricted cubic splines showed a positive dose-response association between sCLEC2 and CAD incidence ( <0.001). The addition of sCLEC-2 to conventional risk factors improved the C statistic (0.821 vs. 0.761, = 0.004) and reclassification ability (net reclassification improvement: 57.45%, <0.001; integrated discrimination improvement: 8.27%, <0.001) for CAD. In conclusion, high plasma sCLEC-2 is independently associated with CAD risk, and the prognostic value of sCLEC-2 may be evaluated in future prospective studies.

Water-soluble BODIPY and aza-BODIPY dyes: synthetic progress and applications

摘要: In recent years, boron-dipyrromethene (BODIPY) and boron-azadipyrromethene (aza-BODIPY) dyes have attracted considerable multidisciplinary attention due to their diverse applications. By introducing various hydrophilic groups, such as quaternary ammonium, sulfonate or oligo-ethyleneglycol moieties into the BODIPY core, the solubilities of these dyes in aqueous solution can be greatly improved while maintaining their high fluorescence quantum yields. Accordingly, applying these fluorescent dyes in aqueous systems to areas such as chemosensors, biomacromolecule labeling, bio-imaging and photodynamic therapy has been achieved. In this article, the recent progress on the synthesis, optical properties and application of water-soluble BODIPY dyes and aza-BODIPY dyes is reviewed.

Synthesis and properties of water-soluble 1,9-dialkyl-substituted BF

摘要: Bis-alkylsulfonic acid and polyethylene glycol (PEG)-substituted BF azadipyrromethenes have been synthesized by an adaptable and versatile route. Only four synthetic stages were required to produce the penultimate fluorophore compounds, containing either two alcohol or two terminal alkyne substituents. The final synthetic step introduced either sulfonic acid or polyethylene glycol groups to impart aqueous solubility. Sulfonic acid groups were introduced by reaction of the bis-alcohol-substituted fluorophore with sulfur trioxide, and a double Cu(I)-catalyzed cycloaddition reaction between the bis-alkyne fluorophore and methoxypolyethylene glycol azide yielded a neutral bis-pegylated derivative. Both fluorophores exhibited excellent near-infrared (NIR) photophysical properties in methanol and aqueous solutions. Live cell microscopy imaging revealed efficient uptake and intracellular labelling of cells for both fluorophores. Their simple synthesis, with potential for last-step structural modifications, makes the present NIR-active azadipyrromethene derivatives potentially useful as NIR fluorescence imaging probes for live cells.

Polymeric ionic liquid—assisted polymerization for soluble polyaniline nanofibers

摘要: To enhance the solubility of polyanilines (PANI), polymeric ionic liquid (PIL) was introduced into the polymerization synthesis of PANI with various proportions. The structure and properties of the modified PANIs were characterized by H NMR, Fourier transform infrared spectroscopy, thermogravimetric analysis, ultraviolet-visible spectrum, etc. It was found that the obtained PANIs doped with PILs were soluble in various organic solvents such as , -dimethyl formamide and acetonitrile. Compared with the pure PANI, the PANIs doped by PILs showed remarkable solubility and their chemical structure and conductivity kept integrated.

Blockage of receptor-interacting protein 2 expression by small interfering RNA in murine macrophages

摘要: This study aims to demonstrate that blocking the receptor-interacting protein2 (Rip2) expression can decrease inflammatory cytokine production by macrophage and protect mice from endotoxin lethality. Murine Rip2 small interfering RNA (siRNA) plasmids were constructed and transfected into macrophage and Rip2 expression was detected with reverse transcription-polymerase chain reaction (RT-PCR) and western blot. Cell proliferation was assayed with MTT. TNF-? concentration was assayed with ELISA and high-mobility group box 1 protein (HMGB1) level with semi-quantitative western blot after lipopolysaccharide (LPS) stimulation. LPS challenge was given after the plasmids were injected into mice and the survival rate was calculated. Rip2 siRNA plasmid could block the mRNA and protein expression of Rip2 and promote cell proliferation. Blocking Rip2 could attenuate LPS-induced TNF-? and HMGB1 production. The HMGB1 expression in the liver decreased to (40.21 ± 11.03) pg/g, and serum TNF-? level decreased to (300.43 ± 59.26) ng/L ( < 0.05). The survival rate of mice from endotoxemia was also improved ( < 0.05). The results demonstrate that Rip2 siRNA plasmid can block the expression of Rip2, decrease the production of TNF-? and HMGB1 and protect mice from fatal endotoxemia.

MSI/LOH and extron expression of the FHIT gene in gastric carcinoma

摘要: We detected loss of heterozygosity (LOH) and microsatellite instabilities (MSI), as well as extron expression of the fragile histidine triad (FHIT) gene in gastric carcinoma (GC), in order to evaluate their association with clinicopathological processes in gastric carcinogenesis. LOH and MSI of the FHIT were detected by using PCR at 4 microsatellite loci: D3S 1300, D3S 4103, D3S 1481, D3S 1234 in cancer tissues from 50 patients with primary GC, with normal mucosa acting as matched controls. FHIT transcripts were detected by nested RT-PCR in 30 cases of GC and their products were sequenced. Results show that the average frequencies of LOH and MSI of the FHIT gene in GC were 32.4% and 26.4%, respectively. There was no correlation between LOH and MSI of the FHIT gene in GC and the histological characteristics of gastric carcinoma (Bormann s or Lauren s classification). LOH of the FHIT gene in GC was related to depth invasiveness, and its frequency in GC where serosa was penetrated was significantly higher than that in GC without serosa penetration (73.5% 37.5%, <0.05). The frequency of MSI in GC without lymph node metastasis was significantly higher than that in GC with lymph node metastasis (66.7% 34.3%, <0.05). Aberrant transcripts were found in 11/30 GC tissues. Sequencing analysis of the aberrant fragments found a RT-PCR product missing exons 5 7 in one case of GC, and another product missing exons 4 7. Four of 10 (40.0%) cases of primary GC showed absent or decreased expression of the FHIT protein as compared to their matched normal tissues. The findings in this study suggest that LOH and MSI of FHIT gene may induce aberrant extron expression, which might play a role in gastric carcinogenesis.
